A petrified Julia Stoner goes to see Sherlock Holmes early in the morningafter having heard a strange whistling sound several times, which she heard once before when her sister died violently and mysteriously of unknown causes,only managing to say the strange words "the speckled band" before she died. She describes her life and the strange circumstances of her sister's death,and that she currently lives with her stepfather Dr Grimesby Roylott and is engaged. Holmes notes that he is an aggressive man, of which he and Dr Watson have personal experience when he bursts into their rooms after Miss Stoner has left, threatening and angry at the thought of Holmes interfering. His instincts aroused that Miss Stoner is in danger, Holmes and Watson go down to Surrey to investigate. They hide in her bedroom and when they hear the whistling noise Holmes lashes out at something, after which there is a scream. Rushing into Dr Roylott's adjoining room they find him dead, bitten by a deadly snake - the "speckled band" - which had turned back on him after being frightened by Holmes. Holmes uncovers that Dr Roylott murdered Miss Stoner's sister when she became engaged, and planned to do the same with her, to cover up the fact that he had used their deceased mother's money which should have gone to them on their marriages.
